Jan Pfeiffer 2023 Abstract The subject of this work is black, or the designation for color. But it is also an artistic concept and means of expression. The subject of my interest are selected cases of the black phenomenon in visual arts, culture, and their possibilities in education. I understand black as a phenomenon that I explore through my own creative work. In the course of it, I ask myself questions, such as: what stimuli, for example, black gives us and how it is connected with our socio-cultural background and its perception. I also ask contemporary artists working consciously or unconsciously with black, what black enables them in their work, and how it supports them, whether it is crucial for them. My intention is to open new perspectives on the subject of black and its connection with daily life, the ability to treat this phenomenon as a conscious gesture, an opinion - distinguishing different uses of black context. Chess Server

This work deals with the creating, and solving the problem of a chess server. It is about creating a chess server wherewith people can play chess through the Internet or a local network and also on a PC. In the real world the construction of the informational system resembles chess games between two players. According to the basic rules of chess the control of the right movements, the analysis of special movements like taking a figure and also rooking, projection and the analysis of checks and mates on the king are solved. During the game statistics of the movements are kept and after the end of the game the performance of the players are calculated by the virtue of record and statistics are kept about the gained ELO (rating) points.
The analysis of cancer invasion plasticity in a 3D environment

iii Abstract Cells have evolved multiple mechanisms of cellular motility ranging from the migration of large cell cohorts to specialized migration of individual cells. The wide range of invasion modes has been exploited by cancer cells to their advantage, which has rendered the metastatic process so difficult to defeat. To allow for a better understanding of cancer invasion plasticity, we have employed studies on cancer cells that adopt the proteolytically active, adhesion-dependent, elongated mesenchymal invasion mode, the protease-independent, low adhesion, rounded amoeboid invasion mode, or combination of both. To study invasion plasticity directly, we have established two model systems of the mesenchymal- amoeboid transition (MAT) that allow for regulated induction of MAT in 3D in vitro environments. Using these systems, MAT was induced in HT1080 fibrosarcoma cells and the acquisition of a motile, invasive amoeboid phenotype was confirmed. We then observed the mesenchymal and amoeboid invasion strategies within 3D collagen in more detail using a digital holographic microscope. The analysis of plasticity of cancer cell invasiveness

The ability of cancer cells to adopt various invasive modes (the plasticity of cancer cell invasiveness) represents a significant obstacle in the treatment of cancer metastasis. Cancer invasiveness involves various modes of migration. Cells can move together (with the preserved intercellular junctions; collective invasiveness) or individually. Within individual invasiveness, we distinguish two principal invasive modes - mesenchymal and amoeboid. The mesenchymal mode of migration is characterized by an elongated shape, proteolytic degradation of the fibres of the extracellular matrix, and the formation of strong contacts with the extracellular matrix. The amoeboid mode of migration is not dependent on proteolytic activity, the cells are characterized by a round shape and increased contractility, which they use to squeeze themselves through the pores of the extracellular matrix. This thesis deals with the analysis of the plasticity of cancer cell invasiveness, specifically the transitions between individual amoeboid and mesenchymal migration modes, in the 3D environment of the collagen gel as a model of extracellular matrix. The plasticity of melanoma cell invasiveness

and keywords: During metastasis, cancer cells can invade the extracellular matrix using various strategies. When invading individually, they employ either the amoeboid invasion mode, during which the cell body dynamically deforms by enhanced contractility to squeeze through pores within the matrix, or protease dependent mesenchymal migration that takes advantage of the possibility to digest the surrounding matrix. Cells migrating in one mode can actively switch to the other by mesenchymal-amoeboid (MAT) or amoeboid-mesenchymal transitions (AMT). This enables escape mechanisms and considerably complicates anti-metastatic treatment. It is well known that Rho GTPases are master regulators of cytoskeleton re-arrangements and thus, unsurprisingly, play a major role in both invasion modes and can directly drive the transitions. However, upstream activation of these pathways is still largely unclear. Word formation of verbs in Russian mat

In this thesis we analyze a linguistic phenomenon in russian language (and East Slavic area) called mat. The task of mat is actual and wide spread in Russian language, but also overlooked in contemporary linguistic scientific literature. The mat is in our thesis analyzed on background of so called typology of swearing, which is a distribution of languages of various language families of european languages based on the common features (themes or functions) of their swearing. In this thesis is also clarified the term mat, it's function and it's composition. The aim of this thesis is word formation process of verbs in mat and it's potential to express non obscenne maenig by using a stem of obscene words, i.e mat. The fourth part compares swearing- vulgarism in Russian and in Czech language, where the focus is to the verbs according to the theme of this thesis.

Diploma thesis has an aim to present to students of elementary school graphical methods of relief printing by method of methodological video, for better understanding and imagination of students. The theoretical part occupies with graphic, the fenomena, that accompanies us from prehistoric times. The thesis is going to present method of relief printing, its development and important authors of relief printing. It makes the students familiar with the most important graphic works in history and makes their analysis. It presents the most common subjects and social aspects of graphical production in the final part. The practical part elaborates technology of instructive video and following application for students of various age and its final marking their work. The conclusion of thesis create supplements of the practical part, examples of models and final works.
